I'm not sure, but I would suspect a corrupted Settings file. Try moving
the Eudora Settings file to a safe location (so you can put it back if
that turns out not to be the problem), then try opening Eudora using the
Eudora Settings.bkup file.
Let us know what happens.

Doesn't solve the problem. Any other ideas? Many thanks in advance

Where is your Eudora (mail) folder, and can you create new files in it?
Web search suggests that "-5000" was formerly
(Mac OS 9 and earlier):
File Manager Extensions Errors
accessDenied - Incorrect access for this file/folder
http://support.apple.com/kb/HT2237
Some Apple mailing list post suggested looking for codes in "MacErrors.h" file.
Or here? http://mac.softpedia.com/get/System-...s-Errors.shtml

Re: Can't create mailboxes
My Eudora Mail folder inside Eudora folder inside documents. And, yes, I
cannot create new files inside the Mail Folder. I don't seem to have
permission to do so. Nor can I move the Mail Folder files to another and
replace the current one.
Help?

So that explains the error encountered by Eudora,
which similarly can't create files there for you.
I'm afraid someone else will have to help you
with fixing the issue.

Select your Mail Folder and do a "Get Info" (Command-I) on it. Check
the "Sharing & Permissions" section and the Privileges listed there.
You should be able to read and write to the folder.

I'd try running Disk Utility to Repair Permissions.

And if that doesn't work, use the Get Info window in the Finder to
change the permissions on the Mail Folder.
