Your best bet is to start with Dreamweaver Help (F1). Under the Contents
tab: Laying Out Pages > Managing Templates > Creating a Dreamweaver template

There is no better way to start and you can be confident you will be doing
things the right way. Otherwise you may mangle the Template and create
endless problems for yourself.

The others have answered your question. Be aware that bringing a site with
pages with existing content (i.e., a retrofit of an existing site) under
template control can be a tedious experience. The only way to do it without
triggering other issues is to follow this procedure:

1. Create your template and save it with FILE | Save As Template.
2. Define the editable regions in the Template file you just saved, and
save it again (FILE | Save).
3. Spawn a child page from the template (with FILE | New From Template >
site name > template name > Create).
4. Open an existing page, and copy/paste content from the page you just
opened to the new child page you just spawned. When done, close the
existing page without saving.
5. Save the newly created child page over the existing page (i.e., use the
same name).
6. Repeat steps 3, 4, and 5 until you are complete.
